TheTopoisomeraseIVDrugScreeningKitcontainsreagentsnecessarytoidentifycompoundsthataffectenzymeactivity. NotethatthiskitdoesnotincludeE.colitopoisomeraseIVenyzme,whichissoldseparately. Thiskitwillallowdetectionoftwokindsoftopoisomeraseinhibitors:thosethatstimulateformationofcleavablecomplexesandthosethatantagonizetopoisomerase(topo)IVactionontheDNA. SincetopoIVisatypeIIenzyme,wemayrefertoitaseithertopoIVortopoII. AknowntopoIIpoisonisincludedasacontrol. Theassaysystemisbaseduponincubatingsupercoiledplasmid(pHOT1)withTopoIVandevaluatingtheformationofDNAcleavageproductswhichmaybenicked,opencircularDNAorlinearDNA. Theproductsarethenresolvedbyethidiumbromidegelelectrophoresisasdescribedinthekit.
